Application
Nuclease from Staphylococcus aureus has been used in a study to assess coagulase and heat-resistant strains found in animals. It has also been used in a study to investigate the expression characteristic of two genes in S. aureus that encode two thermostable nucleases.
Biochem/physiol Actions
Cleavage of DNA by Nuclease micrococcal is greater at the 5′ sides of A or T nucleotides than at G or C.
Hydrolyzes 5′-phosphodiester bonds of DNA.
Other Notes
Note: One μmolar unit = approx. 85 A260 units, where an A260 unit is equivalent to a ΔA260 of 1.0 in 30 min at pH 8.8 at 37 °C in a 3.55 mL reaction volume. Light path = 1 cm.
One unit will produce 1.0 μmole of acid soluble polynucleotides from native DNA per min at pH 8.8 at 37 °C, based on EM/260 = 10,000 for the mixed nucleotides.
