Recognizes the ~130 kDa phosphorylated JAK1 in LIF-treated 3T3-L1 cells. Does not cross-react with other JAKs or related kinases.

      Note: 1 T = 1 test. Sufficient for 10 Western miniblots.
      Catalogue Number420092
      Brand Family Calbiochem®
      Application Data
Detection of mouse JAK1, phosphorylated on Tyr1022/1023, by immunoblotting. Samples: Whole cell lysate from 3T3L1 cells left unstimulated (lane 1) or stimulated with LIF (lanes 2-5). Primary antibody: PhosphoDetect™ Anti-JAK1 (pTyr1022/1023) Rabbit pAb (Cat. No. 420092) (1:1000) pre-incubated without peptide (lanes 1,2), with non-phosphopeptide corresponding to the immunogen (lane 3), a generic phosphotyrosine-containing peptide (lane 4), or phosphopeptide immunogen (lane 5). Detection: chemiluminescence.

      Key Applications Immunoblotting (Western Blotting)
      Application NotesImmunoblotting (1:1000)
Application CommentsA ~110 kDa degradation product is also visible in some cell or tissue preparations. Does not cross-react with any other JAK proteins. Immunogen sequence is conserved in pig JAK1, but cross-reactivity has not been tested. When used at a 1:1000 dilution, there is sufficient antibody for 10 immunoblots at 10 ml/blot. Variables associated with assay conditions will dictate the optimal dilution.

DescriptionRabbit polyclonal antibody adsorbed with non-phosphopeptide corresponding to the immunogen phosphorylation site, followed by immunoaffinity chromatography. Recognizes the ~130 kDa JAK1 protein phosphorylated on Tyr1022/1023.
BackgroundIn response to cytokines or related factors (interferons, interleukins, etc.), JAKs are activated via phosphorylation at 2 adjacent tyrosine residues, as well as perhaps one or more serine residues. Activation of JAKs can lead to the phosphorylation of STAT (signal transducers and activators of transcription) proteins, which dimerize and translocate to the nucleus. JAK1 is required for signal transduction through IL-2, IL-4, IL-6, GM-CSF, and interferon receptors.
