For immunoblotting, incubate with antibody diluted in 1x TBS, 0.1% Tween®-20 detergent, and 5% dry milk overnight at 4°C with gentle shaking. Variables associated with assay conditions will dictate the proper working dilution.

      DescriptionRabbit polyclonal antibody purified by protein A followed by immunoaffinity chromatography. Recognizes the ~58 kDa Smad2 protein phosphorylated at Ser465/467.
      BackgroundThe MAD-related (MADR) family of proteins are intracellular signaling components for the TGFβ superfamily of ligands. Smad2 is activated by phosphorylation by the TGFβ/activin receptor. Activated Smad2 forms a heterocomplex with Smad4 that is then translocated into the nucleus, where it associates with other transcription factors and regulates expression of ligand-responsive genes. Both Smad2 and its closely related homolog Smad3 act as latent nuclear transcriptional activators and mediate intracellular signaling by TGF-betas and activin.
