      DescriptionPurified mouse monoclonal antibody generated by immunizing Balb/c mice with the specified immunogen and fusing splenocytes with p3-NS-1/Ag4-1 mouse myeloma cells. Recognizes the ~42 kDa Mcl-1 protein.
      BackgroundMcl-1 also known as myeloid cell leukemia-1 was originally identified as an early gene expressed in ML-1 cells during phorbol ester induced differentiation along the monocyte/macrophage pathway. Mcl-1 is a member of the bcl-2 family of apoptosis markers and is the anti-apoptotic homologue of bcl-2. The C-terminal regions of Mcl-1 and bcl-2 share significant sequence homology. Mcl-1 together with bcl-2 and the pro-apoptotic protein, bax are thought to function by forming homo- and heterotypic dimers that are critical in controlling the apoptotic process. Like bcl-2, Mcl-1 can be found in epithelial cells in a variety of tissues including prostate, breast, endometrium, epidermis, stomach, intestine colon and respiratory tract although with different expression patterns. Interestingly, in contrast to bcl-2, little or no Mcl-1 is detected in neurons of the brain and spinal cord however, Mcl-1 is highly expressed in cardiac and skeletal muscle which contain relatively less bcl-2. Chondrocytes and hepatocytes do not express bcl-2 but do express Mcl-1. Expression of Mcl-1 is increased upon exposure of ML-1 cells to various types of DNA damaging agents (e.g., ionizing radiation, ultraviolet radiation, and alkylating drugs) along with increases in GADD45 and bax and a decrease in bcl-2. Enhanced expression of Mcl-1 prominently associated with mitochondria complements the continued expression of bcl-2 in ML-1 cells undergoing differentiation. Like bcl-2, Mcl-1 has the capacity to promote cell viability under conditions that otherwise cause apoptosis.
