Background InformationTargeting protein for Xklp2 (Tpx2) is a major spindle assembly factor belonging to the TPX2 family of proteins. Tpx2 is a required component in the normal assembly of mitotic spindles and, during apoptosis, for microtubules. It is also necessary to the nucleation of microtubules in a chromatin and/or kinetechore dependent manner. Tpx2 activates Aurora-A kinase through promotion of autophosphorylation, protects the activated kinase from dephosphorylation, and regulates its localization to spindle microtubules. Normal expression of Tpx2 is exclusive to proliferating cells, specifically those transitioning from G1/S to the termination of cytokinesis. Tissue expression has only been observed in lung cancer cell lines. Phosphorylation of Tpx2 occurs on serine residues; however, the purpose of the modification has yet to be determined.

UniProt SummaryFUNCTION: Spindle assembly factor. Required for normal assembly of mitotic spindles. Required for normal assembly of microtubules during apoptosis. Required for chromatin and/or kinetochore dependent microtubule nucleation. Mediates AURKA localization to spindle microtubules. Activates AURKA by promoting its autophosphorylation at 'Thr-288' and protects this residue against dephosphorylation.

      SUBUNIT STRUCTURE: Interacts with AURKA.

SUBCELLULAR LOCATION: Nucleus. Cytoplasm › cytoskeleton › spindle. Cytoplasm › cytoskeleton › spindle pole. Note: During mitosis it is strictly associated with the spindle pole and with the mitotic spindle, whereas during S and G2, it is diffusely distributed throughout the nucleus. Is released from the nucleus in apoptotic cells and is detected on apoptotic microtubules. TISSUE SPECIFICTY: Expressed in lung carcinoma cell lines but not in normal lung tissues.

DEVELOPMENTAL STAGE: Exclusively expressed in proliferating cells from the transition G1/S until the end of cytokinesis.

PTM: Phosphorylated upon DNA damage, probably by ATM or ATR.

SEQUENCE SIMILARITIES: Belongs to the TPX2 family.
      Molecular Weight~ 95 kDa observed
