Genomic DNA purification from blood in 15 minutes without centrifugation

The MagPrep® Blood Genomic DNA Kit is designed for rapid, small-scale isolation of genomic DNA from blood leukocytes. The method is based on capture of blood cells (leukocytes) on magnetic silica particles, lysis of captured cells, and immobilization of large genomic DNA molecules on the surface of the particles (Cook 1984, Leadon 1982). The resulting DNA:magnetic particles complex can either be used directly in PCR protocols, or the DNA can be eluted before use in PCR by brief warming and mechanical agitation. The protocol yields 0.3–0.6 µg genomic DNA from 10 µl of fresh human whole blood within 15 minutes, and is easy to automate; the separation steps are performed with a magnet and no centrifugation is used. As little as 0.2 µl of the 50-µl elution volume can be used for PCR amplifications. Each kit provides sufficient components for 100 genomic DNA extractions from 10 µl of whole blood.
