The 23 amino acid Cdc25C (182-204) sequence, containing PLK1 (polo-like kinase 1) phosphorylation sites Ser191 and Ser198, is modified between Ser191 and Ser198 as well as after Ser198 according to an optimized concensus sequence derived from 18 PLK1 substrate site sequences. PLK1/2/3 exhibit stronger preference (≥6-fold) for the second serine residue and phosphorylate PLKtide much more potently (≥10-fold) when compared with the parent Cdc25C (182-204) peptide, although PLKtide is not a substrate for PLK4.
