The Single dA™ Tailing Kit is used to prepare
DNA with any type of end for cloning into the
Novagen AccepTor™ Vectors. The kit provides
the reagents needed to blunt DNA ends and
add a single 3′-dA residue. This treatment
creates vector-compatible overhangs and
effectively prevents insert-to-insert ligation,
thus eliminating tandem inserts. The positive
control included in the kit is the blunt-ended
HSV•Tag® 36-mer that encodes the
11-aa peptide recognized by the HSV•Tag
Monoclonal Antibody.
Applications
- Treatment of PCR products of proofreading DNA polymerases for cloning into AccepTor Vectors (alternative to blunt-end cloning)
- Treatment of randomly cut or sheared DNA to be cloned into dU- or dT-tailed vectors
