The LIC cloning procedure uses T4 DNA Polymerase to produce specific non-complementary 12–15 base single stranded overhangs. The proper conversion of the insert ends for ligation-independent cloning requires a specific balance between the exonuclease and polymerase activities of T4 DNA Polymerase. Novagen's T4 DNA Polymerase has been qualified for ligation-independent cloning using a unique assay to verify its performance.
