Sarcolipin (UniProt: O00631; also known as SLN) is encoded by the SLN gene (Gene ID: 6588) in human. Sarcolipin is a single-pass membrane protein that contains a cytoplasmic domain (aa 1-7), a transmembrane domain (aa 8-26), and a lumenal domain (aa 27-31). SLN is a regulator of sarco/endoplasmic reticulum Ca2+-ATPase (SERCA) activity in muscle and plays a role in regulating muscle thermogenesis and metabolism. It increases the heat generated by SERCA by partially uncoupling Ca2+ re-sequestration from ATP hydrolysis in muscle and contributes to non-shivering thermogenesis. This heat generating process is mechanistically distinct from and unrelated to Ca2+-linked thermogenic processes in skeletal muscle. It acts by reducing the apparent affinity of the ATPase for Ca2+. SLN is also reported to modulate calcium re-uptake during muscle relaxation and play an important role in calcium homeostasis in muscle. Higher levels of SLN have been reported in patients with Duchenne muscular dystrophy and reducing SLN levels improves dystrophic pathology. (Ref.: Campbell, KL., and Dicke, AA (2018). Front. Sarcoplasmic reticulum Ca(2+)-ATPases are transmembrane proteins that catalyze the ATP-dependent transport of Ca(2+) from the cytosol into the lumen of the sarcoplasmic reticulum in muscle cells. This gene encodes a small proteolipid that regulates several sarcoplasmic reticulum Ca(2+)-ATPases. The transmembrane protein interacts with Ca(2+)-ATPases and reduces the accumulation of Ca(2+) in the sarcoplasmic reticulum without affecting the rate of ATP hydrolysis.

Studies in sarcolipin knockout mice have led to the suggestion that skeletal muscle sarcolipin plays a role in thermogenesis. The mechanism proposed is uncoupling of the sarcoplasmic reticulum calcium pump. However, in other work sarcolipin was not detected in mouse skeletal tissue. We have therefore measured sarcolipin levels in mouse skeletal muscle using semi-quantitative western blotting and synthetic mouse sarcolipin. Sarcolipin levels were so low that it is unlikely that knocking out sarcolipin would have a measurable effect on thermogenesis by SERCA. In addition, overexpression of neither wild type nor FLAG-tagged variants of mouse sarcolipin in transgenic mice had any major significant effects on body mass, energy expenditure, even when mice were fed on a high fat diet.
