Biochem/physiol Actions

L′anticorps reconnaît tant les formes natives que dénaturées et réduites des protéines de fusion avec une étiquette HA. Il réagit avec les protéines de fusion exprimées dans E. coli ou dans les cellules de mammifères et dont l′étiquette HA se situe à l′extrémité N-terminale ou C-terminale.

General description

L'étiquette HA est une étiquette de 9 acides aminés de séquence YPYDVPDYA (N-Tyr-Pro-Tyr-Asp-Val-Pro-Asp-Tyr-Ala-C) dérivée de la protéine hémagglutinine (HA) au niveau des résidus 98 à 106 de sa séquence d'acides aminés. L'étiquette HA contient une forte proportion de résidus d'acides aminés chargés, ce qui la prédispose à former un puissant site de reconnaissance d'anticorps. Cet anticorps monoclonal anti-HA conjugué à l'agarose reconnaît les formes natives ainsi que les formes dénaturées/réduites des protéines à étiquette HA. Ce produit réagit avec les protéines de fusion portant une étiquette HA à leur extrémité N-terminale ou C-terminale, exprimées dans E. coli ou dans des cellules de mammifères.

Immunogen

Peptide synthétique correspondant à un fragment de l′hémagglutinine (HA) du virus de la grippe humain, connu sous le nom d′étiquette HA ou HA-tag, conjugué à KLH.

Physical form

Suspension dans une solution saline tamponnée au phosphate 0,01 M, contenant de l′azoture de sodium à 15 mM.

Preparation Note

Produit préparé avec de l'agarose activé au bromure de cyanogène. La concentration en anticorps spécifiques varie de 2,0 à 2,4 mg par millilitre de résine tassée.
