Application

La chondroïtinase ABC proposée par Sigma a été utilisée pour digérer des glycosaminoglycanes de type sulfate de chondroïtine (CSGAG) à liaison O. Cette enzyme a également été utilisée pour digérer les chaînes glycosaminoglycanes des protéoglycanes lors de la conception et de l′expression d′un gène de versicane recombinant dans des systèmes de mammifère. Elle a également été employée pour digérer le sulfate de chondroïtine dans les complexes de protéoglycanes Cette enzyme a aussi été utilisée pour éliminer les chaînes de glycane de l′aggrécane, supprimant ainsi son effet inhibiteur sur la croissance de l′axone.

Biochem/physiol Actions

La chondroïtinase ABC catalyse la dégradation par élimination des polysaccharides contenant des liaisons (1-4)-β-D-hexosaminyle et (1-3)-β-D-glucuronosyle (ou (1-3)-α-L-iduronosyle) en disaccharides contenant des groupes 4-déoxy-β-D-gluc-4-enuronosyle. Elle agit sur le 4-sulfate de chondroïtine, le 6-sulfate de chondroïtine et le sulfate de dermatane. Cependant, elle n′agit que lentement sur l′hyaluronate. Son poids moléculaire est d′environ 120 kDa et elle est formée de 2 sous-unités non identiques d′un poids moléculaire de 86 kDa et de 32 kDa. Son pH optimal est de 8,0 pour le sulfate de chondroïtine et 6,8 pour l′acide hyaluronique. Sa température optimale est de 37 °C. Le Zn2+ à 1 mM agit comme inhibiteur de cette enzyme, tandis que l′acétate à 0,05 M est un activateur.

Une unité libère 1,0 μmole d′un mélange de 2-acétamido-2-désoxy-3-O-(acide β-D-gluc-4-ène-pyranosyluronique)-4-O-sulfo-D-galactose et 1,0 μmole de 2-acétamido-2-désoxy-3-O-(acide β-D-gluc-4-ène-pyranosyluronique)-6-O-sulfo-D-galactose à partir de sulfate de chondroïtine dérivée de cartilage de requin, par min à pH 8,0 et 37 °C.

Preparation Note

Reconstituer dans une solution aqueuse comportant de l′albumine de sérum bovin (ABS) à 0,01 %. Des dilutions supplémentaires peuvent être réalisées dans un tampon contenant 50 M de Tris, pH 8,0, avec 60 mM d′acétate de sodium et 0,02 % d′albumine de sérum bovin. Les solutions doivent être fraîchement préparées.

    Chondroitinase ABC is a lyase with a reported molecular weight of approximately 120 kDa by gel filtration and is composed of two subunits with molecular weights of 86 and 32.

    This product consists of approximately 35% protein as determined by the Biuret method. BSA (bovine serum albumin) is a major component (~90%) of the protein. The remaining 65% is Tris buffer salts.

    Reconstitute the product in a 0.01% bovine serum albumin aqueous solution. Subsequent dilutions can be made into a buffer containing 50 mM Tris, pH 8.0, with 60 mM sodium acetate and 0.02% bovine serum albumin. Solutions should be prepared fresh.
