Application

La solution saline tamponnée au phosphate de Dulbecco (DPBS) a été utilisée :

  • pour la dilution de globules rouges frais afin d'étudier le piégeage d'agrégats de globules rouges.
  • pour laver les puits d'une plaque de microtitration.
  • pour laver des cellules avant de les remettre en suspension dans un tampon de lyse pour isoler de l'ADN.
  • comme constituant d'un milieu de conservation pour embryons destiné à la vitrification des embryons.

General description

La solution saline tamponnée au phosphate de Dulbecco (DPBS) est une solution saline équilibrée (BSS, Balanced Salt Solution) utilisée pour la manipulation et la culture des cellules de mammifère. Le tampon DPBS est utilisé pour irriguer, laver et diluer les cellules de mammifère tout en maintenant l'équilibre osmotique. Il apporte l'eau et les ions inorganiques essentiels au métabolisme normal des cellules. Le DPBS additionné de calcium et de magnésium facilite la liaison et l'agrégation cellulaires. Le PBS de Dulbecco sans ajout de ces ions peut être utilisé pour laver et rincer des cellules en suspension. Le tamponnage au phosphate maintient le pH dans la plage physiologique.

  8. What is a Salt Toxicity Test?

    The Cell Culture assay is called a Salt Toxicity Test (previously called Cell Culture Salt Testing). A more complete description is below:Cells are harvested and counted.  They are resuspended in DPBS and incubated at 37°C, 5% CO2 for 1 hour.  The cells are counted and %viability determined.  The cells are then cultured for an additional 7 hours (total of 8 hours).  The cells are counted again and the % viability determined.  The viability at 1 hr has to be >95% of the original viability.  The viability at 8 hrs must be >75% of the original viability.
