General description
Le milieu RPMI 1640 a été mis au point par Moore et ses collaborateurs, au Roswell Park Memorial Institute (RPMI), en 1966. Version modifiée du milieu 5A de McCoy, il a été formulé pour permettre la culture de cellules lymphoblastoïdes en suspension, mais on sait depuis qu'il convient aussi à une grande diversité de cellules adhérentes. Initialement conçu pour être utilisé avec un supplément à base de sérum, le milieu RPMI 1640 s'est révélé adapté à la culture sans sérum de différentes lignées cellulaires. Il est aussi couramment employé dans les protocoles de fusion ainsi que pour la croissance de cellules hybrides. Ce milieu convient à la culture de leucocytes normaux et néoplasiques humains.
Application
RPMI-1640 Medium is used for the following applications:
- Used in cell culture medium: Murine thymoma EL-4 cells were cultured in RPMI 1640 medium supplemented with other components
- Used for the mycobacterial infection of splenocytes (in antibiotic-free RPMI 1640 medium)
- Used for cell isolation and culture
- Used in culture medium (as one of the component) during lymphocyte separation and culture, ELISPOT Assay, Ex Vivo Proliferation Assay
- Used in medium for gp39 gene expression and CD40-Immunoglobuin binding assays
- Used for the preparation of antifungal agents
Preparation Note
Formulated to contain 10.4 grams of powder per liter of medium.
Supplement with 2.0 g/L sodium bicarbonate.
Other Notes
Phenol red has been shown to interfere with the growth of some cells at clonal densities. Use this medium when working with stem cells or when growing cells at low densities. Also recommended for in vitro diagnostics use.
