      DescriptionProtein A purified rabbit polyclonal antibody. Recognizes the ~130 kDa Apaf-1 protein.
      BackgroundThe Apaf-1 (apoptotic peptidase activating factor) gene encodes a cytoplasmic protein that initiates apoptosis. This protein contains several copies of the WD-40 domain, a caspase recruitment domain (CARD), and an ATPase domain. Upon binding cytochrome c and dATP, this protein forms an oligomeric apoptosome. The apoptosome binds to and cleaves caspase-9 proenzyme, releasing mature, activated caspase-9. Activated caspase-9 stimulates the downstream caspase cascade that commits the cell to apoptosis. Alternative splicing of Apaf-1 results in several transcriptional variants encoding different isoforms. Apaf-1 is the ortholog of Ced4 in worms. Alterations in apoptosome function have been documented in a number of tumor types. Modulation of apoptosome function in cancer has been observed in metastatic melanomas in which Apaf-1 protein was lost due to methylation-induced transcriptional silencing.
