      BackgroundSeparase (accession no. NP_036423) is a ~220 kDa cysteine protease essential for both mitosis and meiosis. Cancer cells often have abnormal chromosome numbers, indicating that abberant chromosomal division contributes to cancer development. During cell division, the sister chromatids are held together by the protein, cohesin. During anaphase, separase cleaves cohesin, releasing the sister chromatids. Separse is also essential during meiosis I exit, which is not dependent on the proteolytic activity of separase, but its phosphorylation at Ser1126. Phosphorylated separase forms a complex with Cdk1, which inhibits both the protease and kinase activities, respectively, of the two components, and signals the end of meiosis I.
