      BackgroundHepsin (Acc # NP-002142.1) is a type II transmembrane serine protease that belongs to a family of related proteases that are grouped into four subfamilies. The subfamilies includeh hepsin/transmemebrane proteinase, serine (TMPRSS); matriptase; corin; and the human airway trypsin (HAT)/DESC subfamilies. Hepsin contains a hydrophobic region of 27 amino acids located at the N-terminus, serving as a transmembrane signal, and an extracellular C-terminal domain with serine protease activity. Hepsin is upregulated in advanced stages of prostate, ovarian, and renal cell carcinomas. Hepsin can activate coagulation factor VII, suggesting that it may also play a role in blood coagulation. In addition, hepsin efficiently activates the proform of hepatocyte growth factor (HGF), a multifunctional growth factor that acts through its high-affinity receptor tyrosine kinase, c-Met, and is tightly regulated by HGFA inhibitor 2 (HAI-2). Antibodies that neutralize hepsin activity have been shown to inhibit the invasive properties of some prostrate and ovarian tumor cells.
