      Background InformationInterleukin-15 (UniProt: P40933; also known as IL-15) is encoded by the IL-15 gene (Gene ID: 3600) in human. IL-15 is a cytokine that is predominantly regulated post-transcriptionally at the levels of translation and intracellular trafficking. It stimulates the proliferation of T-lymphocytes and it shares many of the biological properties of IL-2, including stimulation of CTLL proliferation, and in vitro generation of alloantigen-specific cytotoxic T cells and nonantigen-specific lymphokine-activated killer cell. Stimulation by IL-15 requires its interaction with components of the IL-2 receptor, including IL2RB and probably IL2RG but not IL2RA. One of the most critical roles of IL-15 is in the development, survival and activation of natural killer cells. IL-15 is synthesized with a signal peptide (aa 1-29) and a propeptide (aa 30-48) that are subsequently cleaved off to generate the mature form (aa 49-162). Two isoforms of IL-15 have been described that are produced by alternative splicing. Isoform IL15-S48AA is an extracellular secreted form and isoform IL15-S21AA is not secreted but is stored intracellularly in the nucleus and cytoplasmic components. IL-15 is shown to be most abundant in placenta and skeletal muscle and is also detected in heart, lung, liver, and kidney. Isoform IL15-S21AA is preferentially expressed in testis and thymus.
