      BackgroundSmac, also known as DIABLO, is a mitochondrial protein that promotes cytochrome c-dependent caspase activation by neutralizing inhibitor of apoptosis proteins (IAPs). Smac functions at the levels of both the Apaf-1-caspase-9 apoptosome and effector caspases. Smac is translocated to the cytosol during apoptosis. Mitochondrial import and cleavage of its signal peptide are required for Smac to gain its apoptotic activity. Processing of Smac to the mature form is independent of apoptosis.
