      Kinase Activity Assay
      Required Materials • Assay Buffer: 50 mM Tris-HCl, 10 mM NaCl, 10 mM DTT, 0.05% Brij®-35 detergent, pH 7.5 • Phosphatidylserine (PS): 10 mg/ml PS in chloroform • Phosphatidylcholine (PC): 10 mg/ml PC in chloroform • Phosphatidylethanolamine (PE): 10 mg/ml PE in chloroform • Phosphatidylinositol (PI): 1 mg/ml PI in chloroform • PI 3-Kα, p110α/p85α: 0.33 mg/ml, diluted to 0.01-0.05 mg/ml in Assay Buffer • ATP/MgCl2 Mix: 5 µM ATP, 100 mM MgCl2 • EasyLite™ Kinase Reagent: available from Perkin Elmer • 96-well plate Preparation of Reagents • Lipid Mix: for each assay, in a glass vial, mix 0.5 µl PC, 1.2 µl PS, 1.1 µl PE, 1.5 µl PI. Let it stand for ~30 min until the chloroform is evaporated. Reconstitute with 30 &µl Assay Buffer without Brij®-35 detergent. Activity Assay 1. To designated wells, add 30 µl Lipid Mix and 10 µl PI 3-Kα (100-500 ng/assay). Incubate for 5 min at 22°C. 2. Add 10 µl ATP/MgCl2 mix and incubate for 30 min at 22°C. 3. Add 50 µl pre-warmed EasyLite™ Kinase Reagent. 4. Measure activity using a luminometer.
