      Application NotesDILUTION:

      Optimal dilution is to be determined by the user.

      PROCEDURE USED FOR QUALITY CONTROL TESTING OF CLONES:

      1. Remove the acetone-fixed Positive and Negative Control impression slides from the freezer and allow to equilibrate to room temperature.

      2. If uncoated slides are used, encircle each impression with an ImmuPen or wax pencil.

      3. Add the diluted reagent directly onto control and test slides. Care must be taken to avoid cross-contamination of slides during the staining procedure.

      4. Incubate in a humid chamber at 37°C for 30 minutes.

      5. Remove excess antibody from the slides by briefly rinsing with PBS. Next, wash the slides for 10 minutes with PBS. Change the PBS once during this time.

      6. Rinse briefly with deionized water and allow to air dry.

      7. Mount coverslip using mounting fluid containing 10% glycerol at pH 8.5.

      8. Examine the mounted slides using a fluorescence microscope. Slides not examined within the working day may be stored at 2 -8°C for 24 hours. If longer storage is required, slides should be stored at -20°C.
