Full-length, recombinant, human UbcH3 fused at the N-terminus to His•Tag® sequence and expressed in E. coli. UbcH3, an E2 ubiquitin conjugase homologous to Cdc34 in yeast, is important in the control of cell cycle and DNA replication. UbcH3/Cdc34 in association with different E3 complexes, including SCF, has been shown to target proteins including IκB, B-Myb, Wee1, MyoD, ICERIIγ, ATF5, p27Xic1, and p27Kip1. Other targets such as β-catenin, p21Cip1, E2F, cyclin E, and cyclin D are putative substrates of Cdc34 by virtue of their SCF requirement for proteolysis. The active enzyme is useful in the study of ubiquitin regulation and inhibitor screening.
