      A cell-permeable pyrazolo-pyrimidine compound that acts as a potent ATP-binding pocket-targeting inhibitor against p38α MAP kinase activity (IC50 = 5 nM; Ki = 0.2 nM), while exhibiting little or no activity against a panel of 21 other commonly studied kinases and 5 CYP 450 isozymes (IC50 ≥40 µM). Shown to be orally active in mice with excellent pharmacokinetics and in vivo efficacy in lowering (>70%; 5 mg/kg p.o. 5 h prior to LPS challenge) LPS-induced plasma TNFα production.
