c-Raf-1 は Ras のエフェクターとして機能し、入ってくる信号をミトゲン活性化タンパク質キナーゼ経路に伝達するセリン-スレオニンキナーゼです。この経路は、細胞の増殖、生存、および分化を含むさまざまな細胞応答に関連しています。RBD は、in vitro で Ras·GTP 複合体に直接結合します。さらに、細菌で発現された組み換え RBD は、in vitro で安定した折り畳まれた構造をとります。セファロースビーズに結合した GST·RBD 融合タンパク質は、GTP と恒常的に結合している発癌性 Leu61-Ha-Ras を沈殿させるために使用できます。

Raf-1 RBD、GST 300 μg、0.5 mM PMSFおよび20%グリセロールを含有するPBS（pH 7.4）150 μLに溶解。

活性化Rasのアフィニティー沈降： Raf-1-RBD GSTをグルタチオンコーティングプレートに捕捉し、これを用いてHeLa細胞ライセート由来のGTP活性化Rasを捕捉することができます。 1ウェルあたり2 μgのRaf-1-RBD、GSTを用いて、25～100 μgのEGF刺激HeLa細胞ライセートを捕捉しました。 Upstate社は、Ras GTPase ELISAキット（17-424）も提供しています。
注：Rasは自然に失活するため、-70℃での保存期間が6ヵ月未満のライセートを使用してください。
