Application

この分子試薬はポリアニオン色素であり、カチオン性のクエンチャーDPX(X1525)と組み合わされることが多く、補体媒介性の溶解など、膜融合や膜透過性のアッセイに使用されます。タリウムイオンとセシウムイオンにより、効率的に蛍光が抑制されることが確認されています。神経細胞トレーサーとして使用されています。本製品は水でのストークスシフトが比較的大きく、生物学的サンプルのほとんどの自己蛍光の発光から十分に分離します。D2Oにより本製品の蛍光量子収量が4倍上昇し、これは赤血球ゴーストや腎集合管の水透過性の判定に有用であることが確認されています。炭水化物の還元末端はANTS分子の一級アミノ基と反応してシッフ塩基を形成し、シッフ塩基はシアノ水素化ホウ素ナトリウムにより還元されて二級アミンになります。この還元的アミン化反応は酸触媒であるため、反応液のpHにより、動態やANTSによる炭水化物の誘導体化の程度が変化することがあります。注意点として、この反応は中性pHでも進行可能で、オリゴ糖のシアル酸結合は酸加水分解の影響を受けやすいため、シアル酸結合の完全性を保つには中性pHが適しています。そのようにして炭水化物をANTSで標識すると、混合物をキャピラリー電気泳動で分離することができます。
