Biochem/physiol Actions
ペプシンやレニン、カテプシンDおよび多数の微生物アスパラギン酸プロテアーゼを含む酸プロテアーゼの、強力なインヒビターです。有効濃度1 μM。約1日安定。
ペプスタチンAは酸プロテアーゼ(アスパルチルペプチダーゼ)のインヒビターです。ペプスタチンAは、例えばペプシンやレニン、カテプシンD、ウシキモシン、プロテアーゼB(Aspergillus niger)といったプロテアーゼと、1:1複合体を形成します。このインヒビターは非常に選択的で、チオールプロテアーゼや中性プロテアーゼ、またはセリンプロテアーゼを阻害しません。可溶化γ-セクレターゼおよびレトロウイルスプロテアーゼは、ペプスタチンAでも阻害されます。様々な試料由来のプロテアーゼの特性を明らかにするために使用されてきました。
Preparation Note
酢酸とメタノールとの混合液(10% (v/v)、メタノール:酢酸 = 9:1)に可溶であり、溶解性は1 mg/mLです。メタノールまたはDMSOに溶解する際に、酢酸の添加が必要になることがあります。エタノールへの溶解性は1～2 mg/mLですが、完全に溶解させるには加熱が必要になる場合があります。溶液は60°Cまで加熱してもペプチドが分解することはありません。
濃度1 mg/mLのストック溶液は4°Cで1週間は安定です。メタノールまたはDMSOに濃度1 mMで溶解した溶液は-20°Cで数ヶ月間安定です。溶液の黄色が濃くなっている場合は加水分解が起こっています。
使用濃度は1 μMで、室温で1日は安定です。
濃度1 mg/mLのストック溶液は4°Cで1週間は安定です。メタノールまたはDMSOに濃度1 mMで溶解した溶液は-20°Cで数ヶ月間安定です。溶液の黄色が濃くなっている場合は加水分解が起こっています。

1 Uは、ペプシンの活性を1 U(ヘモグロビン(製品番号51290)を基質に用い最終容量16 mLとしてpH 2.0、37°Cで1分間に280 nmの吸光度を0.001増加させる酵素量に相当)低下させるインヒビターの量に相当します。10,000吸光度-ユニットは1 Bergmeyer-ユニット(1ペプシン-ユニットをpH 2.0、37°Cで1分間に1 μmolのアセチル-L-フェニルアラニル-3,5-ジヨード-L-チロシンを加水分解する酵素量とした場合)に相当します。
