General description

ウシ血清アルブミン（BSA）は、ウシ肝臓で生成される球状の非グリコシル化αらせん構造の血漿タンパク質です。3つのドメイン（I、II、およびIII）で構成され、各ドメインは2つのサブドメイン（AおよびB）を有し、ハート型の構造を呈します。

Application

ウシ血清アルブミンは以下に使用されています-

  • BTS（Beltsville thawing溶液）とともに、精子運動解析における精子を伸展するため
  • modified Krebs-Ringerバッファー（dmKRBT）の成分として
  • ゲルシフトアッセイに
  • 免疫細胞染色、免疫組織染色(4)、免疫蛍光プロトコルおよびサンドイッチ酵素結合免疫吸着測定法（ELISA）のブロッキング剤として
  • フローサイトメトリー分析のために細胞を再懸濁するためのリン酸緩衝生理食塩水（PBS）の成分として
  • 組織サンプル中のタンパク質をブラッドフォード法によって定量するためのタンパク質標準として
  • 17β HSD酵素の酵素活性を測定するため

Preparation Note

しばしば、"Cohn Fraction V"と呼ばれます。Cohnの冷エタノール分画法の改良法により調製。
血清アルブミンはフラクション（画分）Vと呼ばれる場合があります。この命名規則は、冷エタノール沈殿を用いて血清タンパク質を分画するオリジナルのCohn（コーン）法から取られています。血清アルブミンは、Cohn法を用いて第5エタノール画分に認められました。それ以来、「フラクションV」という用語は、調製の方法に関係なく血清アルブミンを記述するのに使用されています。他の人はこの用語を使用して、オリジナルのCohn法が記述されて以降大幅に改変されたエタノール分画法により精製された血清アルブミンのことを記述しています。Sigma-Aldrichは、真のCohn分画法、改変エタノール分画法、熱ショックやクロマトグラフィーなど様々な主要方法で精製された血清アルブミンを製造および販売しています。さらなる精製ステップには、結晶化や活性炭ろ過が含まれる場合があります。
