General description

アプロチニンは、分子量の計算値が6512 Daの58アミノ酸-長鎖タンパク質で、セリンプロテアーゼ阻害剤として作用します。ウシ膵臓トリプシン阻害剤（BPTI）としても知られるアプロチニンは、キモトリプシン、カリクレイン、プラスミンおよびトリプシンなどのセリンプロテアーゼを阻害します。

Application

アプロチニンは、トリプシンの阻害剤として主に使用されます。

Biochem/physiol Actions

アプロチニンは競合的セリンプロテアーゼ阻害剤で、酵素と安定な複合体を形成し活性部位をブロックします。この結合は可逆的で、ほとんどのアプロチニン‐プロテアーゼ複合体は pH が 10 を超えるか、または 3 を下回るような極端な pH レベルでは解離します。アプロチニンは、構造的には、58 アミノ酸からなるウシ肺由来のグロブリン様タンパク質モノマーであり、単一のポリペプチド鎖中の 3 つのジスルフィド結合によって架橋されています。

Preparation Note

この製品は、0.9% NaClおよび0.9%ベンジルアルコールを溶媒として、無菌的に充填した溶液です。 この製品はタンパク質1 mg当たり3～7 TIU、溶液1 mL当たり3～21 TIUの活性を有します。

Other Notes

1 トリプシン阻害剤ユニット（TIU）は、2 トリプシン単位の活性を 50％減少させ、1 トリプシン単位は、pH 7.8 および 25°Cにおいて、1 分間で 1 μ モルの N-α-ベンゾイル-DL-アルギニンp-ニトロアニリドを加水分解します。他にも一般的に使用される単位として、KIU があります。1 TIU = 1,300 KIU で換算します。
