General description
LH、FSH、TSHのものと同一の92アミノ酸で構成されるα鎖と、LH、FSH、TSHのものとは異なる149アミノ酸で構成されるβ鎖からなる糖タンパク質ホルモンです。絨毛組織によって産生され、妊娠初期の黄体の維持に関与しています。また、栄養膜腫瘍によっても産生され、遊離hCG β鎖と高度にグリコシル化されたhCGの増加によって特徴づけられます。
Application
絨毛性ゴナドトロピンは、
- 細胞毒性のin vitro分析に
- 卵子および胚調製物に
- CD-1マウスの過剰排卵に
- 自然周期にあるラットを同期化するための黄体形成ホルモン放出ホルモン(LHRH)アゴニスト(LHRH-a)を用いたin vivo研究のためのゴナドトロピン調製物に使用されています。
Biochem/physiol Actions
ヒト絨毛性ゴナドトロピン(hCG)は、実験動物の排卵を誘導するPMSGの代替として使用できます。 hCGを実験動物に静脈内(i.v.)、腹腔内(i.p.)または皮下(s.c.)注射したときの動物に対する50%有効量(ED50)の動物当たり、および体重(kg)当たり(括弧内)の値は以下の通りでした。
マウス: i.v.0.2 (7.7) I.U.; i.p.0.3 (11.5) I.U.; s.c.0.7 (26.9) I.U.

Preparation Note
水で1,000 IU/mLの濃度にもどすと、バイアルは0.1 Mりん酸ナトリウム緩衝液(pH 約7.2)と10 mg/mLのマンニトールも含むことになります。

計算値はヒト絨毛性ゴナドトロピンに対するUSP法に基づいており、バイアル当たり10,000 IU、80.0%以上の忍容性と規定効力の125.0%以下を満たします。
