General description
LH、FSH、TSHのものと同一の92アミノ酸で構成されるα鎖と、LH、FSH、TSHのものとは異なる149アミノ酸で構成されるβ鎖からなる糖タンパク質ホルモンです。絨毛組織によって産生され、妊娠初期の黄体の維持に関与しています。また、栄養膜腫瘍によっても産生され、遊離hCG β鎖と高度にグリコシル化されたhCGの増加によって特徴づけられます。
ヒトの絨毛性ゴナドトロピン（CGAまたはhCG）はヘテロ二量体であり、シスチンノット成長因子スーパーファミリーに属しています。6つのバリアント、すなわちhCGを含むhCGn、hCGβ、hCGβn、hCGβcfおよびhCGαとして存在します。CGAはヒト染色体6q14.3にマッピングされます。hCGはαとβ鎖で構成されています。
Application
ヒトの絨毛性ゴナドトロピンは以下の用途に使用されています。
- 培養卵胞内で卵成熟を誘導するために
- 妊娠中の雌馬血清ゴナドトロピン（PMSG）とともに、マウスの過剰排卵を誘導するために
- ステロイド産生反応性を促進する胎児精巣培養のための培地成分として
Biochem/physiol Actions
トロホブラスト（栄養芽細胞）癌では、hCGレベルの上昇がみられます。 hCGレベルは妊娠の指標であり、βサブユニットは主に尿サンプル中に存在します。hCGアイソフォームの検出は、癌を含むトロホブラスト関連疾患の診断と治療に役立ちます。
Preparation Note
水で1,000 IU/mLの濃度に戻すと、バイアルは0.1 Mりん酸ナトリウム緩衝液（pH約7.2）と100 mg/mLのマンニトールも含むことになります。
Analysis Note
計算値は、ヒト絨毛性ゴナドトロピンのUSPメソッドに基づいており、バイアルあたり5,000 IUを満たし、許容範囲は記載されている効力の80.0%以上、125.0％以下です。
