General description

フィブロネクチン1は、FN1遺伝子によってコードされる、細胞外マトリックスの糖タンパク質です。血漿や細胞表面に発現しています。FN1遺伝子はヒト染色体2q35にマッピングされます。
FNは、2つのジスルフィド結合によってC末端で結合した二量体タンパク質として生じます。フィブロネクチン構造内では、ドメインはペプチド鎖のアミノ末端とカルボキシ末端のいずれにも近い位置で密に配列されています。さらに、単一の遺伝子に由来するにもかかわらず、この遺伝子の異なる変異体がヒトで20個まで同定されたとの報告があります。

Application

ヒト血漿由来のフィブロネクチンは、H₂O₂の摂取速度の測定に使用されています。また、細胞培養用のSylgard^®モデルのコーティングにも使用されています。また、細胞接着や、ポリマーフィルム上へのタンパク質の直接パターニングにも使用されています。

Biochem/physiol Actions

フィブロネクチンの機能には、細胞の接着、増殖、遊走、創傷治癒、血液凝固、および転移などのさまざまな生物学的過程が含まれます。フィブロネクチンタンパク質は、細胞の接着や伸展、細胞骨格の制御、形態学や分化において重要な役割を果たします。FN1はまた、細胞外マトリックスの形成、止血、血栓症にも関与しています。FN1の変異は、糸球体症を引き起こします。

Preparation Note

本製品は、フィブロネクチンの0.1％溶液です。タンパク質の均一性は、免疫電気泳動によって評価されており、また、エンドトキシン試験済みです。原料は、HIV、ならびにB型およびC型肝炎ウイルスに陰性です。

培養用の表面コーティングでは、フィブロネクチンを滅菌緩衝塩溶液で希釈し、できるだけ少ない液量でコーティングしてください。その後、表面を45分間、室温で乾燥させます。2～8°Cで2～4週間保存できます。

タンパク質の均一性は免疫電気泳動法により確認されています。
