General description

フィブロネクチンは、多数のドメインを有する糖タンパク質です。N末端の中央結合ドメインと、ヘパリン結合ドメインとから構成されています。ウシ血漿フィブロネクチンは、ジスルフィド結合によるポリペプチド鎖を有し、分子量は220～215 kDaに相当します。

フィブロネクチンは接着糖タンパク質であり、細胞膜表面タンパク質としても血漿中にも見られます。フィブロネクチン（FN）は、複数の機能を持った糖タンパク質であり、その遺伝子は、ヒトクロモソームの2q34-36に位置します。この遺伝子は全長75 kbで、50のエクソンを含みます。可溶型と不溶型の2つの主なアイソフォームとして存在します。前者は血漿中に存在し、一方、不溶型は組織や軟骨の細胞外マトリックス（ECM）中に存在します。可変領域の選択的スプライシングと、エキストラドメインA、Bと呼ばれる2つのタイプIIIエクソンにより、FNには約20の異なるフォームが存在します。

Biochem/physiol Actions

フィブロネクチンは、タンパク質の足場形成の調節剤です。血小板の各種機能、創傷治癒、接着、移動、凝集などの細胞プロセスを調整します。ヒト歯髄（HDP）細胞は、初期の接着とその後の伸展をフィブロネクチン（FN）に依存することが研究によって示されています。FNはマイトジェン活性を示し、細胞増殖を促進するとともに、歯髄細胞の象牙芽細胞への分化過程の調節に関与している可能性もあります。FNの存在は、特に歯の象牙質形成に関与する象牙芽細胞への分化において、HDP細胞の運命と発生に影響を及ぼすと考えられます。
