糖タンパク質であるヘパラン硫酸プロテオグリカン（HSPG）は、基底膜に不可欠な部分です。HSPGは、ヘパラン硫酸鎖に共有結合したコアタンパク質で構成されます。HSPGには、膜HSPG、分泌型細胞外マトリックスHSPG、および分泌小胞プロテオグリカンという3つのグループがあります。

Application

ヘパラン硫酸プロテオグリカンは、以下に使用されています：

  • リン酸緩衝生理食塩水（PBS）に使用され、調査研究においてトラウマ当量濃度を得るために新鮮クエン酸血液サンプルに添加される
  • 固相結合アッセイ
  • 細胞外マトリックス（ECM）マイクロアレイスライドの作製

Biochem/physiol Actions

ヘパラン硫酸プロテオグリカン（HSPG）は、さまざまな生物学的過程において複数の機能を発揮します。基底膜構造に関与し、また細胞遊走のためのマトリックスを提供します。serglycinなどの分泌小胞におけるHSPGは、顆粒内容物のパッケージング、プロテアーゼ活性の維持、および分泌後の生物学的活性の調節に関与します。HSPGは、サイトカイン、増殖因子、ケモカインに結合してこれらを保護することができ、細胞の特定と白血球の動員に不可欠な勾配の形成を促進します。これらはプロテアーゼおよびプロテアーゼ阻害剤の受容体として作用し、細胞-ECM接着、細胞-細胞相互作用、および細胞運動性を調節し、また増殖因子受容体と連携できます。また、HSPGはリガンドクリアランスのためのエンドサイトーシス受容体としても機能し、リポタンパク質代謝とモルフォゲン濃度勾配の形成に関与します。また、マトリリシンやその他のマトリックスメタロプロテイナーゼのドッキング分子としても作用します。ヘパラン硫酸は、培養中の細胞の付着を促進し、10～100 ng/mLの濃度でヘパリン硫酸および可溶性FGF受容体が欠乏している細胞へのFGF-basicの高親和性結合を誘導します。

Preparation Note

この製品は、pH 7.4の50 mM Tris HCl、150 mM NaCl、1 mM EDTA、および0.1 mM PMSFを含有する溶液中でEngelbreth-Holm-Swarmマウス肉腫の基底膜から単離されます。
