Application
ラクトフェリンは、鉄制限培地におけるStreptococcus mutans（ミュータンス菌）の増殖のために使用されました。ラクトフェリンが、in vitroで上皮細胞の接着を妨げるかどうかを検証するために使用されました。 また、急性肺炎における血中サイトカイン濃度の診断的価値を検証するためにも使用されました。
Biochem/physiol Actions
ラクトフェリンは鉄結合タンパク質です。血漿中の鉄輸送タンパク質であるトランスフェリンと構造が類似していますが、鉄に対する親和性はラクトフェリンの方が顕著に高値です(250倍)。初乳に非常に多量に含まれるほか、涙液、唾液、粘膜分泌物や好中球の二次顆粒中にも存在することがあります。粘膜上皮および好中球によって産生され、炎症性刺激に応答してこれらの細胞から放出されます。鉄を封鎖する能力により細菌の成長を阻害し、またN末端を介してリポ多糖類と結合することで細菌細胞壁を透過性にします。ラクトフェリンはウイルスのエンベロープタンパク質に強く結合するためウイルス感染を阻害できます。結合ドメインをブロックし、細胞とウイルスの融合を防ぎます。ラクトフェリンは宿主の防御系を活性化すると考えられており、この作用の一部は好中球活性化因子であるインターロイキン-8の放出の促進によるものです。抗体およびインターロイキンの合成、リンパ球増殖、補体活性化にも関与していると考えられています。
