Analysis Note

凝集活性は、μg/mLの単位で表され、レクチン、カルシウム、マグネシウム、およびマンガンをそれぞれ異なる濃度で含むリン酸緩衝生理食塩水(pH  6.8)を用いた1 mg/mL溶液を段階希釈して決定します。凝集活性は、25°Cで1時間インキュベートしたときに、適切な赤血球の2%懸濁液を凝集させる最低濃度として示されます。

Application

一般的なウェスタンブロットプロトコル：
  • 糖タンパク質サンプルサイズ：500 ng
  • レクチン濃度： 0.1 ug/mL

  1. レーンあたり500 ngの糖タンパク質をサンプルとして添加します
  2. 4～20% Bis-Tris SDSページゲル電気泳動を実施します
  3. ゲルをPVDFメンブレンに転写します
  4. RIPAバッファー（R0278 Sigma）でRTで1時間メンブレンをブロックします
  5. RTで2時間、RIPAバッファーで0.1 ug/mLのHRPレクチンをインキュベートします
  6. 25 mL RIPAバッファーで膜を5 x 5分間洗浄します
  7. 化学発光基質（CPS1-120）を使用して検出します

Biochem/physiol Actions

BS-Iは、末端のα-D-ガラクトース残基に最も強く、次に末端のN-アセチルα-D-ガラクトース残基に親和性を示します。

Other Notes

1 unitは、pH 6.0、20°C、20秒間に、1 mgのプルプロガリンをピロガロールから生成する酵素量です。
BS-Iは、A・Bと呼ばれる2種類のサブユニットからなる四量体のレクチンです。BS-Iにはサブユニットの組成が異なる5種類のイソレクチンが存在し、それぞれBSI-B4, BSI-AB3, BSI-A2B2, BSI-A3B, BSI-A4と呼ばれます。BSI-B4はB型血液に特異的で末端α-D-ガラクトシル残基だけに親和性を示しますが、一方、BSI-A4はA型血液に特異的で、末端N-アセチル-α-D-ガラクトサミニル残基に主要な親和性を有しています。
