一般的なウェスタンブロッティングのプロトコール：
  • 糖タンパク質サンプル量：500ng
  • レクチン濃度：0.1ug/ml

  1. サンプルを1レーン当たり糖タンパク質500 ngになるようにロードします
  2. 4～20% Bis-Tris SDS PAGEゲルで分離します
  3. ゲルをPVDF膜に移します
  4. RIPAバッファー（R0278 Sigma）を用いて室温で１時間、膜をブロッキングします
  5. HRPレクチンを0.1ug/mlでRIPAバッファーと共に室温で２時間、インキュベートします
  6. 25mlのRIPAバッファーで5分間、膜を洗浄します（5回）
  7. 化学発光基質（CPS1-120）で検出します

Biochem/physiol Actions

PNAは正常ヒト赤血球は凝集しませが、ノイラミニダ-ゼ処理赤血球を強く凝集します。PNAには、ヒト血清中の抗T細胞抗体と同様に、強い抗T細胞活性があります。レクチンはヒトリンパ球のサブセットを識別するのに用いられます。

公表されている方法（低分子量の結合が優先される方法）を変更し、ペルオキシダーゼ（P8375）から調製。結合後、アフィニティークロマトグラフィーにより再精製。
