凝集活性は、1 mg/mL溶液を段階希釈して測定し、mg/mLの単位で表します。凝集活性は、25°Cで1時間インキュベートした場合、ヒト赤血球2%懸濁液(PBS、pH 6.8)を凝集させる最低濃度で示されます。有糸分裂活性は、HeLa細胞培養液中のBrdU取り込み量により決定されます。
Application
Phaseolus vulgaris（赤インゲン豆）由来レクチンは、フィトヘマグルチニン（PHA）アッセイや、細胞免疫および液性免疫の評価に使用されます。
また、in vivo T細胞媒介免疫応答性を測定するために抗原として使用されています。
Biochem/physiol Actions
PHAは、分裂促進活性は低くエリスロアグルチニン活性が高いエリスロアグルチニン（PHA-E）と、分裂促進活性とロイコ凝集活性は高くエリスロ凝集活性が非常に低いロイコアグルチニン（PHA-L）という2つの分子種で構成されています。
PHA-Eは血液型特異的ではなく、凝集反応は特定のオリゴ糖によって阻害されることがあります。PHA-Pは、エリスロアグルチニンとロイコアグルチニンを分離・精製する前のたんぱく質形態のPHAです。PHA-Mはムコたんぱく形態です。共役は対応する精製レクチンから調製されます。

フィトヘマグルチニン（PHA）は、植物（特にマメ科）に認められるレクチンです。Phaseolus vulgaris（赤インゲン豆）から得られ、ヒト培養白血球においてマイトジェン活性を示します。
