この製品は、0.5～1.0 mLの0.1 mg/mL溶液を使用して25 cm2をコーティングする場合の細胞培養基材として推奨されます。より低い分子量バージョンの製品は粘性が低くなりますが、より高い分子量バージョンの製品は、分子あたりの接着部位が多くなります。

Biochem/physiol Actions

ポリ-D-リジン（PDL）臭化水素酸塩は、細胞用の非特異的付着因子であり、細胞膜の負に帯電したイオンと培養物表面との間の静電相互作用を高めることにより、固体基質への細胞接着を促進するのに役立ちます。 ポリ-D-リジンは、培養物表面への吸収後、正に帯電した細胞結合部位の数を増加させます。

Preparation Note

HBrを除去するには、本製品を中性の緩衝液に溶解し、塩を透析除去します。一般に、本製品を結合因子として使用するには、無菌組織培養グレードの水50 mLを5 mgのポリリジンに加え、25 cm2当たり1 mLの溶液で表面を無菌的にコーティングします。5分後、吸引によって溶液を取り出し、表面をよく洗います。細胞と培地の導入前に2時間乾燥させます。

Other Notes

ポリ-D-リジンは、1つのリジン残基あたり約1つのHBrを有する正に帯電したアミノ酸重合体です。ヒドロブロミド（臭化水素酸塩）は、ポリ-D-リジンが水に溶ける結晶形になることを可能にします。HBrはアミノ基と、カルボキシル基またはNまたはO含有部分のいずれかとの間の水素結合を妨げるので、少量の生成物がß構造に認められる場合があります。

Disclaimer

滅菌溶液は、2 ～ 8°Cで保存した場合、最大 2 年間安定です。-20°Cでは乾燥させて保存してください。
