MN551 is an E3 ligase suppressor of cytokine signaling 2 (SOCS2) inhibitor that blocks SOCS2 substrate binding by covalently modifying SOCS2 SH2 domain EF loop Cys111. MN551 also effectively modifies SH2 containing protein (CISH) (Cys114), but not SOCS4 (Cys350) or SOCS6 (no EF loop Cys). Preincubation of K562 lysate with MN551 (1.5h at r.t.) blocks endogenous SOCS2 from binding immobilized GHR pY595 substrate peptide (IC50 = 5.9 µM).
