ββ-1チューブリンは、微小管の主要な構成成分であるチューブリンのサブファミリーに属しています。ββ-チューブリンの分子量は55 kDaです。β−チューブリンの構造は、a-ヘリックスに囲まれた2つのβ-シートで構成されるコアを特徴としています。グアニンヌクレオチド結合領域を伴うN末端ドメイン、タキソール結合部位を伴う中間ドメイン、分子モータータンパク質の結合表面を含むC末端ドメインも有しています。ヒトβ-チューブリンは、7つのアイソフォーム（βI（クラスI）、βII（クラスII）、βIII（クラスIII）、βIVa（クラスIVa）、βIVb（クラスIVb）、βV（クラスV）、およびβVI（クラスVI））で構成されています。
モノクローナル抗β-チューブリン抗体（マウスIgG1アイソタイプ）は、マウス骨髄腫細胞とウニ（Lytechinus pictus）精子軸糸タンパク質で免疫化したBALB/cマウスの脾細胞の融合により産生されるD66ハイブリドーマから得られます。

Application

マウスで産生されたモノクローナル抗β-チューブリン抗体は、ウエスタンブロット法や免疫細胞化学で使用されています。
透過処理精子モデルにおける鞭毛運動の阻害を必要とする試験に使用できます。
マウスで産生されたモノクローナル抗β-チューブリン抗体は、免疫蛍光アッセイと酵素結合免疫吸着アッセイ（ELISA）にも使用されています。

Biochem/physiol Actions

マウス脳β-チュ-ブリンアイソフォ-ムのすべて(βおよびβ′亜群)とウニβ2-チュ-ブリンを認識します。抗体により認識されるエピト-プは、β-チュ-ブリンのC末端領域(ウニβ2-チュ-ブリンのアミノ酸427-432)に存在しています。
遺伝子の変異により、滑脳症、厚脳回症、多小脳回奇形などのさまざまな神経細胞移動異常症がもたらされます。B2702ペプチドはβ-チューブリンに結合してナチュラル・キラー（NK）細胞の細胞毒性を阻害するとともに、細胞骨格の組織化とチューブリンのシャペロン様活性を損なう微小管重合に影響を与えます。ββ-1チューブリン（別名クラスVIまたはTUBB1）は、血小板産生に極めて重要な役割を担い、免疫性血小板減少症（ITP）バイオマーカーの有望な候補と考えられています。
