General description

αチューブリンα 4a（TUBA4A）とも呼ばれるα-チューブリンは、ヒトクロモソーム 2q35 にマッピングされています。この遺伝子は、448 アミノ酸を含む、α-チューブリンファミリーのメンバーをコードしています。TUBA4A は、微小管の主要な構成成分であるチューブリンのサブファミリーに属しています。αチューブリンのα-サブユニットの分子量は 50,000 です。

Biochem/physiol Actions

アセチル化チューブリンは、様々な生物における軸糸の集合に関係しています。アセチル化チューブリンは、微小管の構造や機能の区別において重要な役割を果たしている可能性があります。α-チューブリンの翻訳後修飾、つまり、特異的なアセチラーゼによる Lys40 へのアセチル基の可逆的付加により、アセチル化チューブリンが生成します。
抗体はChlamydomonas軸糸α-チュ-ブリンのα3アイソフォ-ムに存在するアセチル化されたLys-40を含む4残基内のエピト-プを認識します。抗体で検出されるChlamydomonasの配列とほぼ同一の配列が大部分のα-チュ-ブリンに認められますが、相当する領域はニワトリ、Drosophila、酵母などある種のα-チュ-ブリンアイソフォ-ムでは明らかに異なっています。この抗体は、複数の生物に存在するアセチル化α-チュ-ブリンの検出に用いられています。抗体により認識されるエピト-プは、ネズミカンガル-の上皮性細胞株PtK2のように存在しないか、あるいはマスキングされている場合があります。
