General description

モノクローナル抗チロシンチューブリン抗体（マウスIgG3アイソタイプ）は、マウス骨髄腫細胞と免疫化マウスの脾細胞を融合して作製したハイブリドーマに由来します。円筒状フィラメント構造体であるチューブリンは、ほぼすべての真核生物細胞に存在します。チューブリンは、α-チューブリンとβ-チューブリンで構成されるヘテロ二量体です。これらのサブユニットは共に分子量が55 kDaであり、高い相同性を有します。
細胞内の円筒状フィラメント構造である微小管はチューブリンを主成分とし、すべての真核生物細胞に存在します。モノクローナル抗チロシン抗体は、間接的免疫蛍光標識法によるチロシン化α-チューブリンの免疫細胞化学的局在証明に使用できます。モノクローナル抗チロシン抗体は、ウシ脳、サバンナモンキー腎細胞（Vero）、イヌ腎（MDCK）、有袋類腎（Potoroo、PtK2）、マウス下垂体腫瘍（AtT-20）、酵母、アフリカツメガエルのチロシンチューブリンと特異的に反応します。

α-チュ-ブリンのC末端アミノ酸を含むペプチド。

Application

モノクローナル抗チロシンチューブリン抗体は、次の用途で使用されています：
  • 間接的免疫蛍光標識
  • 免疫ブロッティング法
  • 免疫細胞化学的染色

Biochem/physiol Actions

チューブリンは、微小管の主要な構成成分です。チューブリンのチロシン化は、チューブリンの会合状態に関与します。特異的チューブリニルチロシンカルボキシペプチダーゼは、末端チロシンを除去し、グルタミン酸残基を末端とするα-チューブリンを生成します。もうひとつの酵素はカルボキシ終端にチロシンを付加することでα-チューブリンを修飾します。これによりチロシン付加とチロシン削除のサイクルが生じると思われます。
本抗体は、イムノブロッティングにおいて、チュ-ブリンC末端チロシンに結合します。培養細胞や組織切片においても、このエピト-プを検出できます。
