General description
MAPT（微小管関連タンパク質タウ）としても知られているタウ（τ）は、ヒト染色体17q21.3にマップされた遺伝子によってコードされています。ニューロンで高度に発現されますが、軸索で最も顕著です。
タウは、神経細胞の微小管の安定性および構造を制御すると考えられている微小管関連タンパク質のファミリーです。タウタンパク質ファミリーは、単一の遺伝子に由来する代替mRNAスプライシングバリアントに由来し、352～441個のアミノ酸（45～60 kDa）のサイズの異なる成熟タンパク質をもたらします。タウは、微小管結合活性を喪失しており、神経変性障害において対らせん状細線維（PHF）に凝集します。PHFは、神経原線維変化（NFT）の基本的な構造成分です。NFTの蓄積は、アルツハイマー病の臨床的進行と相関しています。リン酸化はタウの機能特性に影響を与える可能性があり、タウの過剰リン酸化はタウの微小管結合活性の喪失と不溶性凝集体の形成をもたらす可能性があります。過剰リン酸化および非酵素的グリコシル化はPHFタウで検出される翻訳後修飾であり、正常タウおよびPHFタウともに多数の過剰リン酸化部位が確認されています。

タウ（τ）は微小管構造の構築と維持に不可欠な役割を果たしています。タウ（τ）が欠失すると、発達遅延と学習障害が生じます。本遺伝子の発現はアルツハイマー病（AD）の発症に関連しています。τ遺伝子の遺伝的変異は、孤発性タウオパチー、進行性核上性麻痺（PSP）、大脳皮質基底核変性症に対する感受性のリスクを高めます。
マウスモノクローナルクローンTau46抗タウ抗体は、ウシ、ラット、ヒト、およびマウスのタウを認識します（約45～60 kDa）。アミノ酸404～441個（ヒト）のリン酸化非依存性エピトープを認識します。タウの6つのアイソフォーム全てを認識し、MAP2タンパク質と交差反応する可能性があります。
