Analysis Note

Le pH optimal se situe entre 2 et 4. Activité maintenue dans une solution d′urée à 4 M et de guanidine HCl à 3 M. Stable à 60 °C. La pepsine est inactivée de façon irréversible à pH 8,0 - 8,5.

Application

La pepsine est une peptidase utilisée pour digérer les protéines et est souvent utilisée pour la préparation des fragments Fab d′anticorps. La pepsine dérivée de la muqueuse gastrique porcine a été utilisée pour l′hydrolyse d′échantillons cervicaux secs chez la souris. Le produit P7000 est fourni sous forme de poudre et a été utilisé pour traiter les cellules épithéliales provenant d′un carcinome mammaire unique félin.
La pepsine peut être utilisée pour produire des fragments F(ab′)2 d′anticorps. pepsine sur www.sigma-aldrich.com/enzymeexplorer.
L′enzyme proposée par Sigma a été utilisée pour la stimulation in vitro de la digestion gastrique du cabillaud cuit. Elle a été utilisée pour la stimulation in vitro de la digestion gastrique de la masse de cacao et des fibres alimentaires en supplément. Elle a également été employée pour augmenter la fraction de collagène soluble extractible et pour réduire l′immunogénicité du collagène qui en découle dans le tissu dermique de bovin.

Biochem/physiol Actions

Clivage préférentiel : résidus hydrophobes et aromatiques en positions P1 et P1′. Clive les liaisons Phe-Val, Gln-His, Glu-Ala, Ala-Leu, Leu-Tyr, Tyr-Leu, Gly-Phe, Phe-Phe et Phe-Tyr dans la chaîne β d′insuline.
La pepsine hydrolyse les liaisons peptidiques, mais pas les liaisons amide ou ester. La pepsine clive les peptides contenant un acide aminé aromatique d′un côté ou l′autre de la liaison peptidique. Les acides aminés contenant du soufre augmentent la susceptibilité à l′hydrolyse lorsqu′ils sont à proximité de la liaison peptidique. La pepsine clive préférentiellement du côté carboxyle des résidus de phénylalanine et de leucine et du côté carboxyle des résidus d′acide glutamique. Clive les liaisons Phe-Val, Gln-His, Glu-Ala, Ala-Leu, Leu-Tyr, Tyr-Leu, Gly-Phe, Phe-Phe et Phe-Tyr dans la chaîne β d′insuline
La pepsine est l′enzyme protéolytique principale produite dans l′estomac. Elle digère les protéines en clivant les liaisons peptidiques internes.
L′enzyme ne réalise pas de clivage au niveau des liaisons avec la valine, l′alanine ou la glycine. La Z-L-tyrosyl-L-phénylalanine, la Z-L-glutamyl-L-tyrosine et la Z-L-méthionyl-L-tyrosine peuvent servir de substrats à la digestion par la pepsine. La pepsine est inhibée par plusieurs peptides contenant de la phénylalanine.

  3. I've seen the term "1:10000" used to define Product P7000, Pepsin from porcine gastric mucosa  (Pepsin A). What does that mean?

    A historical measurement of pepsin activity was the ratio of pepsin to coagulated egg white that the pepsin would digest. Thus 1:10,000 would refer to one part pepsin to 10,000 parts coagulated egg white.  More recent analyses use hemoglobin as a substrate, in place of coagulated egg white.

  4. Can I freeze stock solutions of Product P7000, Pepsin from porcine gastric mucosa  (Pepsin A)?

    Solutions of the enzyme at pH 4.4 should be stable at -20 °C for 2-3 months.

  5. Product P7000, Pepsin from porcine gastric mucosa  (Pepsin A), can be stored at room temperature, but other pepsins are stored in the cooler or freezer.  Why is this?

    This relates to autodigestion of pepsin.  P7000 is a relatively "crude" pepsin, where other proteins that are present would be more likely digestion targets.  With more purified pepsin, the risk of autodigestion increases, and storage at colder temperatures reduces that risk.
