Application

テトラサイクリンは広汎なスペクトルを持つポリケチド抗生物質で、ロッキー山発疹熱、発疹チフス、ダニ熱、Q熱、およびブリル‐ジンサー病などの細菌感染の治療、および上気道感染およびニキビの治療のために臨床的に使用されます。多剤耐性や急性膵炎を含む重篤な副作用の研究に使用されています。細胞培養では、10 mg/Lでの使用が推奨されています。

Biochem/physiol Actions

作用機序：テトラサイクリンは受動的に細胞膜中のタンパク質チャネルを通して拡散し、30Sリボソームに結合し、mRNA-リボソーム複合体上の受容体部位へのアミノアシルtRNAのアクセスを妨げることにより、タンパク質合成を阻害します。またテトラサイクリンは、細菌の50Sリボソームサブユニットに結合し、膜を変化させ、菌体からの細胞内成分の漏出も引き起こします。阻害効果が洗浄により回復可能であることから、テトラサイクリンは可逆的に結合する抗生物質であり、非可逆的に結合する薬剤ではないこと、これが抗菌作用の原因であることが示唆されます。

耐性の機序：細胞壁の浸透性が消失することで効果は不活性化されます。

抗菌性スペクトル：グラム陽性およびグラム陰性細菌に対し広範囲な抗菌活性があります。

Preparation Note

本製品は、水に溶けやすく、メタノールおよびエタノールに可溶ですが、エーテルおよび炭化水素には不溶です。本製品は、水中で加温すると、透明な黄色～オレンジ色の溶液となり、95%エタノール4 mLに50 mgを溶解し加熱すると、透明な黄色～緑色の溶液となります。テトラサイクリンは、水酸化アルカリ溶液によってすぐに分解し、水溶液中で放置すると加水分解や沈殿によって濁ります。テトラサイクリンの活性は、pH2以下の溶液では低下します。
