過ヨウ素酸で処理すると、グリコールがアルデヒドに酸化されます。シッフ試薬(パラローズアニリンとメタ重亜硫酸ナトリウムの混合物)との反応後、 パラローズアニリン付加物が放出され、これがグリコールを含有する細胞成分をピンク～赤色に染色します。PAS法によって呈色する細胞成分は、グリコーゲン、真菌壁、基底膜、特定の上皮スルホムチンとシアロムチン、中性粘液物質、甲状腺の膠様物質、下垂体中間部です。
