Application

DL-ジチオスレイトールは、血小板細胞質調製にも使用される緩衝液の調製に使用される極めて重要な成分です。また、グアニジウム塩を用いたRNAの単離および組織と細胞の均質化においても重要です。
SH基を還元状態に保つ試薬です。ジスルフィドを定量的 に還元します。SDS-PAGEのサンプルバッファーに加えることで、タンパク質のジスルフィド結合の還元に効果を発揮します。また、架橋剤であるN,N′-ビス(アクリロイル)シスタミンのジスルフィド結合を還元してポリアクリルアミドゲルのマトリックスを切り離す目的でも用いられます。2-メルカプトエタノールと比較して、刺激性および毒性が抑えられています。通常、2-メルカプトエタノールの使用濃度(5% v/v、700 mM)の7分の1濃度(100 mM)で効果を発揮します。

Biochem/physiol Actions

DL-ジチオスレイトールは、ジスルフィド結合を還元し、それらをSH基に還元することによってタンパク質を変性させます。また、タンパク質のシステイン残基間の、分子内および分子間ジスルフィド結合の形成も防止します。
