アムホテリシンBは、ストレプトマイセス属由来のポリエン系抗真菌抗生剤です。ステロール、特に、真菌と細菌の両方の細胞膜の成分であるエルゴステロールに高い親和性を示します。細胞培養の用途で培地中にペニシリンやストレプトマイシンとともに使用する場合の用量は、通常2.5 mg/Lです。

Biochem/physiol Actions

アムホテリシン B はステロールと結合して、膜内に細孔を形成し、低分子の漏れ出しを引き起こします。本製品は、真菌や酵母に対して最も効果的です。

Disclaimer

アムホテリシンBは、37℃での培養で3日間活性を維持します。空気と光から保護し、-20℃で保管した場合、本製品は5年間活性を維持します。

Preparation Note

アムホテリシンBは、pH6～7の水には溶けませんが、pH2またはpH11の水には溶けます。DMSO（30～40 mg/mL）およびジメチルホルムアミド（2～4 mg/mL）に溶けます。溶解度が低いため、水溶液は滅菌ろ過できません。
