ブタ膵臓から分離されるα-アミラーゼは糖タンパク質です。残基数475個未満の単一ポリペプチド鎖であり、SH基2個およびジスルフィド架橋4個を有し、Ca2+が強く結合して安定化に寄与しています。塩素イオンは活性と安定に必要とされています。活性のpH範囲は5.5～8.0であり、至適pHは7です。

Application

α-アミラーゼは、スターチ、グリコーゲンなどのα結合ポリサッカライドのα結合を加水分解するために使用されます。 Sigmaが提供するα-アミラーゼは、シロイヌナズナの代謝試験などさまざまな植物研究に使用されています。 ブタ膵臓由来のα-アミラーゼは、トマトにおけるスターチ代謝および熟成の研究でスターチを除去するために使用されています。

Biochem/physiol Actions

α-アミラーゼは、3個以上のα-（1.4）結合D-グルコース単位から成るポリサッカライド中のα-（1.4）グリカン結合を加水分解します。 スターチ、グリコーゲンなどの天然基質は、グルコースとマルトースに分解されます。 ブタ膵臓由来のα-アミラーゼは、2SH基を含む約475個の残基、4個のジスルフィドブリッジおよび安定のために必要な強く結合したCa2+1個を有するポリペプチド単鎖で構成される糖タンパク質です。 最適pHは7です。
