General description

Taq DNAポリメラーゼは好熱菌Thermus aquaticus由来の熱安定性酵素です。E. coli（大腸菌）で発現される組換え型酵素です。95℃までの反復加熱に対し、活性を大きく損なうことなく耐えることができます。Taq DNAポリメラーゼの各ロットは、PCR増幅および二本鎖シーケンシングについて検査されています。

Application

Thermus aquaticus由来のTaq DNAポリメラーゼは、以下の用途に使用されています：
  • ポリメラーゼ連鎖反応（PCR）および光度アッセイによる真菌増殖の定量に
  • 従来の逆転写（RT）-PCRに
  • 単純反復配列（SSR）遺伝子型決定に

Biochem/physiol Actions

Taqポリメラーゼは、オリゴヌクレオチドプライマーによる、DNAテンプレート依存的な、相補的DNA鎖へのdNTPの取り込みを触媒します。本酵素は、5'から3'のポリメラーゼおよびエキソヌクレアーゼ活性の両方を示します。

Taq DNA polymerase は、高温細菌Thermus aquaticusから分離された専用の熱安定性酵素です。大腸菌で発現したリコンビナント酵素です。SDS-PAGEで検出したところ、この94kDaのタンパク質にエンドヌクレア-ゼ・エキソヌクレア-ゼのコンタミネ-ションは認められません。この酵素は、5′→3′ポリメラ-ゼ活性とエキソヌクレア-ゼ活性の両方を有しています。
